Why is the や smaller in one of the examples? Is it simple variation, or is there a difference between the two?

The definition is the same. As I understand it, the small ゃ can be used only when the last character of the verb stem is one that can be combined with the small character. Example: かる/わかる - Stem is わかり. 'りゃ' is a valid combination, so it can be written as わかりゃしない.
